Do you mean a cover letter or a query letter?

The letter one sends an agent to seek representation for getting a novel published is a query letter. The agent's submission guidelines would be a factor.

A cover letter might be used by a freelancer for submitting articles, short stories, or poetry to a publisher, and would be influenced by that particular publisher's submission guidelines.
