Correcting spelling (if you didn't do it on the fly because your editor has a good spell checker) and correcting grammar should be the last things you do, not the first. Why spend time correcting these things, only to dump part of your manuscript (and those corrections) because it is not necessary or needs rewriting?
Look at the large, structural things first, then general things such as characterisation and leave the line-edits until last.
