No. You can only add one Passkey to an item at a time. If you enter a new passkey, that removes the initial passkey. Anyone trying to enter the old passkey will not have access.
Remember, passkeys work in conjunction with the item's "Access Restriction" feature. If an item's Access Restriction is set to "Public", setting the Passkey will have no effect, as the item is publicly accessible to everyone regardless of the Passkey. The most restrictive option is setting Access Restrictions to "Keep Private, for my eyes only!" and adding a passkey.
