Overall Impression
Lovely prose!

Your 'ministry' is showing up a bit in here. I can imagine someone standing up and pontificating. That's not to say a reader was lost among the words, but this definitely has a 'sermon' feeling to it. Just my take.

A few thoughts
"the heart or essence of our humanity - has proven the most enigmatic puzzle of all."
I would counter with 'has proven the most disastrous of all.'

We're killing the planet that supports us. We have the highest brain function, but unfortunately, we're the most destructive. Other than humans, I know of only one other species that kills its own.

Not to be singing 'Doomsday is Tomorrow' because, as you discussed in this essay, we'll never figure out the mystery. What's the point in chasing after golden nuggets? Is it the search itself some crave?

The middle section presents as a large block of text. I think there are places to begin a new paragraph(s). Possibly here:

*Bullet* If I find myself beginning any sentence with 'again' I read what I've written and ask 'is any of this repetitive?'

*Bullet* Every writer has their own style. For me, full capitalization of a title is too showy. Or, like shouting in a quiet room. Reading is a silent pastime.

Lasting Impression
I could sit here all night and go on and on about this topic, but for me, it's not a pleasant intellectual exercise. I find only tragedy in life. Not in a morbid, depressive way, more like an acceptance of whatever happens.

Your last sentence made we wince. The words hurt.

I'm not philosophizing. I have no philosophy. Nor is the quest for answers consuming me. In this little place of mine, let there be happiness and comfort.
